Quantification of quorum sensing components produced by bacterial biofilms

The goal of this research project is to create a method of in situ anal. for quorum sensing mols., N-Butyryl-L-homoserine- lactone and Autoinducer-2 within in Pseudomonas aeruginosa biofilm in real time. In this work N-Butyryl-L-Homoserine-Lactone and Autoinducer-2 will be analyzed for relative recovery from a microdialysis probe using bioluminescence of Vibrio harveyi. Following confirmation of recovery, creation of a feasible method for P. aeruginosa biofilm formation on the semi-permeable membrane of a microdialysis probe will be performed. Once a feasible method has been created, formation of P. aeruginosa biofilms on microdialysis probes will proceed and dialyzate will be collected. Anal. of quorum sensing within a P. aeruginosa biofilm will include monitoring the prodn. of N-Butyryl-L-homoserine-lactone and Autoinducer-2 in real-time. Completion of this project is a necessary prerequisite to integration of microdialysis sampling as a tool to analyze and ultimately redirect quorum sensing during biofilm formation on devices.
