Comparing MALDI and ESI FTMS IRMPD and CID

The infrared multiphoton dissociation (IRMPD) in a Fourier Transform mass spectrometer (FTMS) technique was compared with matrix assisted laser desorption ionization (MALDI) and electrospray ionization (ESI) sources. The FTMS equipment includes both electrospray and MALDI ion sources. All peptides and proteins used in the experiment were procured from commercial sources. A good ESI IRMPD fragmentation pattern was obtained on the protein ubiquitin (8KDa) but not on insulin (5KDa) or cytochrome c (12KDa). The fragments matched calculated m/z values with several charge states for fragmentation.
