Comparison of identified proteins from Escherichia coli whole cells by MALDI TOF and FTMS

The comparison of matrix-assisted laser desorption/ionization (MALDI) time-of-flight (TOF) and fourier transform mass spectrometry (FTMS) methods which were used for the characterization of Escherichia coli (E.Coli) whole cells was discussed. The bacteria which were grown in tryptic soy broth (TSB) were combined into 50 mL centrifuge tubes and spun down at 20000 rpm for twenty minutes, for the characterization. It was observed that the molecular ions in the MALDI-MS spectra were identified by comparing their masses with the mass of E. Coli proteins in the SwissProt/TrEMBLE database. It was observed that FTMS identified more proteins than TOF due to higher resolving power and better mass accuracy.
