Synthetic studies toward antascomicin B

Antascomicin B is a member of a family of macrolides isolated from a strain of Micromonospora that exhibits FKBP12 binding activity as potent as rapamycin and FK506 (IC50 0.7 nMol). We have previously demonstrated in model systems that novel variants of the Ireland-Claisen and allylic diazene rearrangement can be used to install the C23, C26 and C27 stereocenters. Applications of these methods toward the total synthesis of antascomicin B will be described.
