Enhanced microdialysis sampling of antidepressant drugs: Influence of cyclodextrins on protein binding equilibrium.

Microdialysis sampling with inclusion of binding agents such as ss-cyclodextrin (ss-CD) has been used in vitro and in vivo to improve the recovery of hydrophobic compds. However, to date, there have been no theor. or exptl. studies performed to det. if continuous localized infusion of binding agents such as the cyclodextrins affect the binding equil. to proteins such as albumin. Imipramine and desipramine are known to bind ss-CD as well as human serum albumin (HSA). In these studies, in vitro microdialysis sampling was performed with solns. contg. imipramine and desipramine in HSA to compare exptl. relative recovery results with an equil. model. Understanding how the localized binding equil. are affected during enhanced microdialysis sampling with cyclodextrins is an important step towards using this technique for sampling other hydrophobic drugs in vivo.
